Intitulé du projet : Perturbations cutanées induites par l’exposome : recherche de signatures moléculaires et d’effecteurs, exploration d’une approche corrective.
La fonction première de la peau est de constituer une barrière physique entre le corps et l'environnement extérieur, le protégeant ainsi des agents pathogènes, des facteurs toxiques et de la déshydratation. Différents stress de l’exposome peuvent altérer son intégrité, notamment les rayonnements ionisants (radiothérapie) et solaires, entraînant respectivement le développement de fibrose et la mise en place d’une élastose. Ce projet s’intéresse à deux composants de ces processus physiopathologiques : les modifications des fibroblastes dermiques et les désorganisations matricielles affectant la structure et la fonctionnalité du derme.
Un premier versant du projet portera sur la compréhension de la reprogrammation conduisant aux transformations pathologiques du derme. Il s’appuie sur l’accès à des cellules et échantillons de peau humaine représentatifs d’états sains et perturbés. Ce matériel sera mis à profit pour la réalisation de profilages moléculaires ‘multi-Omics’ sur cellules, ainsi que pour des analyses spatiales sur tissus.
Un second versant portera sur l’obtention d’une répression de caractères associés aux désorganisations dermiques. L’approche aura pour principe la vectorisation d’ARNs actifs ciblant des effecteurs de ces désorganisations. L’objectivation fonctionnelle bénéficiera de l’accès à des modèles cellulaires et organoïdes générés à partis de cellules primaires de peau humaine.
Vous travaillerez au sein du Laboratoire de Régénération et Radiopathologies Cutanées’ (LR2C), laboratoire porteur du projet, situé au sein du biocluster Évry-Génopole.

Vos missions
Vous bénéficierez d’un environnement multidisciplinaire pour mener à bien vos missions
Préparation de sous-populations de cellules primaires extraites d’échantillons de peau humaine, sélectionnées selon différents caractères phénotypiques et fonctionnels (présence sur site d’une plate-forme de tri par cytométrie en flux).
Réalisation de profilages moléculaires ‘multi-Omics’ (compétences en bio-statistique et bio-analyse présentes en interne).
Mise en œuvre de modèles fonctionnels ex vivo (cultures 2D et organoïdes cutanés 3D).
Travail de synthèse inhérent au suivi du programme (rapports, réunions)
Rédaction de publications.&lt;br /&gt;&lt;br /&gt;
Votre profil 
Vous êtes titulaire d’une thèse dans le domaine biomédical.
Compétences techniques et comportementales
Compétences en génomique fonctionnelle (RNA-seq, single-cell RNA-seq, ATC-seq, etc.)
Connaissances en biologie cutanée et/ou biomécanique
Culture cellulaire 2D (cellules primaires humaines) et expertise sur organoïdes 3D.
Cytometries en flux, RT-PCR, méthodes d’imagerie.
Travail d'équipe, autonomie, capacité à travailler dans un environnement multidisciplinaire.

Since the COVID-19 pandemic, the CNRGH has also become a key contributor to the Obépine consortium, which aims to develop wastewater-based epidemiological surveillance strategies for infectious diseases. Initially focused on SARS-CoV-2, Obépine now targets the emergence of a broad spectrum of pathogens.
In this context, we are seeking a research engineer to perform the bioinformatics analysis, interpretation of high-throughput sequencing data derived from wastewater samples and implementation of innovative approaches in viral genomics.
The candidate will:
·         Implement and optimize bioinformatics pipelines for the analysis of viral genomes from complex environmental samples, in accordance with the best practices and community guidelines.
·         Process and analyze data from multiple sequencing platforms, including Oxford Nanopore, Illumina, Element Biosciences and PacBio technologies, generated by target enrichment and/or metagenomics approach.
·         Contribute to the development of robust analytical frameworks for wastewater-based surveillance. This includes the reconstruction of viral genomes, variant analysis, haplotype assignation, and the interpretation of metagenomic data from heterogeneous and noisy samples.
·         Collaborate closely with wet lab scientists to integrate experimental design and bioinformatics analysis, to provide reporting solutions to wet lab scientists, and to support the development of innovative and reliable approaches for pathogen monitoring in wastewater.
·         Contribute to conference presentations and scientific articles, and provide a thorough reporting of bioinformatics developments.
Applicants should hold a PhD or an engineering degree in bioinformatics, computational biology, applied mathematics, or a related field, and demonstrate strong experience in one or more of the following: Python, C, C++, R, or bash. Candidates should have a proven experience with next-generation sequencing data analysis, metagenomics and/or viral genomics, and strong basis of pipeline development (such as sequence alignment, genome assembly, variant calling, and metagenomics approaches). Experience in virology, environmental genomics, statistics, or phylogenetics would be a strong advantage, as well as prior experience with high-performance computing clusters.
The position requires the ability to work effectively in a multidisciplinary environment, with strong analytical thinking, scientific rigor, autonomy, and organization skills. A strong interest in continuous scientific and technological monitoring is also expected. Good written and verbal communication skills in French and English are required.&lt;br /&gt;&lt;br /&gt;
