Description de l'unité

The Laboratory of Chemistry and Biology of Metals (LCBM) is a joint research unit (UMR 5249) affiliated with the French Alternative Energies and Atomic Energy Commission (CEA), the French National Centre for Scientific Research (CNRS), and Université Grenoble Alpes (UGA). It is located within the Grenoble Institute for Interdisciplinary Research (IRIG). The scientific research program of the LCBM, at the interface between chemistry and biology, focuses on studying the structure, activity, and regulation of complex biological systems that transport and use metal ions, as well as on leveraging this knowledge to develop bio-inspired systems with similar structures and/or functions. It is a research center dedicated to the study of the “chemistry and biology of metals,” with a unique critical mass and scientific coherence at both the national and international levels. The researchers at the LCBM develop multidisciplinary approaches to address three major societal challenges: the energy transition, reducing environmental impacts, and health. The research themes, which define the laboratory's teams, are Biocatalysis (BioCat); Physical Chemistry of Metals in Biology (PMB); Bioinorganic Catalysis and Environment (BioCE); Proteomics for Microbiology, Immunology and Toxicology (ProMit); Solar Fuels, Hydrogen and Catalysis (SolHyCat); Metals and Organs (MET&OR); Bioenergy & Environment (BioCE); and Computational Chemistry, Modeling and X-rays (CoMX).

The REPAROX project aims to understand and harness repair mechanisms of [Fe-S] enzymes, key biocatalysts in biotechnology whose production and activity are severely limited under aerobic conditions, and translate this mechanism through an ab initio design approach to iron–sulfur biocatalysts of biotechnological interest.

The project builds on the recent discovery of a novel [Fe-S] repair mechanism, involving:

. An intrinsic component linked to the structure of the Quinolinate synthase NADA enzyme

. An extrinsic component associated with the mitochondrial environment

The project combines: Genetics, Molecular biology, Structural biology,  Biochemistry, Spectroscopy, Computational protein design, and engineering metabolic pathways in yeast

IMPACT: REPAROX aims to overcome a major bottleneck in the aerobic bioproduction of [Fe-S] enzymes, enabling the development of more robust and tailor-made biocatalysts for industrial applications.

Within the REPAROX project, the BioCat team in Grenoble aims to biochemically, enzymatically, and spectroscopically characterize NadA from several organisms, both as an isolated enzyme and in complex with the protein partners involved in its repair mechanism under oxidative conditions.

Profil du candidat

Formation : PhD in biochemistry

Compétences : The candidates should have a strong expertise in biochemistry and enzymology and an interest for spectroscopy, structural biology and metals in biology. Candidates should preferably have a 2 to 4 years research experience

Expériences et activités sur place : expression and purification of proteins, enzymatic assay, metal content determination, biophysical characterization (SEC-MALLS, CD, Mössbauer and EPR spectroscopy, BLI (protein-protein interaction)).
